I'm keen on the apparently random look that can be achieved with the 3 sizes of Marshalls Tegula Drivesett (240 x 160; 160 x 160; 120 x 160). Are there formulae for calculating what mix of these 3 sizes works best?

There are. Marshalls have theirs; I have mine, and almost every other contractor in the land has theirs. I don't like to have too many of the TS blocks (120mm), but some folk (perversely) like to use hundreds of the little buggers!
You need to know your total area and then figure out what combination of packs will give you that area plus 5% for wastage. You also need to factor in what blocks would be used for edge courses and adjust your calculations accordingly.
For argument's sake, let's say you worked out that 7 packs of TLs, 3 of TMs and 1 of TSs would give you just what you needed. This does NOT mean a laying ration of 7:3:1 as each pack contains different numbers of blocks....
7 @ 280 = 1960
3 @ 420 = 1260
1 @ 560 = 560
...which is actually 14:9:4
